Camptothecin has been efficiently obtained by a three- or four-component domino Knoevenagel / hetero-Diels-Alder reaction of aldehyde, Meldrum's acid, and enol ether in the presence or absence of alcohol, followed by reductive cleavage of the amine protecting group.
