This is often explained by the depreciation of human capital, the signaling effect of (long) employment breaks and the possible discrimination of mothers in the labour market. However, research has neglected the question what influence it has whether mothers stay out of work beyond the maximal length of the legal leave of absence or not. We expect wage penalties particularly for those mothers who extend their leave of absence. Besides we examine whether the negative impact of employment interruptions varies between mothers with different amounts of human capital in the form of vocational qualifications and between East and West Germany. Our analysis is based on data from the GSOEP (1984-2011) and controls for unobserved heterogeneity. Overall, our results show that there is no general or permanent \"motherhood wage penalty\", but that mainly breaks exceeding the legally suggested duration lead to lower wages, as they probably signal a low career orientation of mothers. The production of the central inhibitory transmitter GABA (gamma -aminobutyric acid) varies in response to different patterns of activity. It therefore seems possible that GABA metabolism can determine inhibitory synaptic strength and that presynaptic GABA content is a regulated parameter for synaptic plasticity. 2. We altered presynaptic GABA metabolism in cultured rat hippocampal slices using pharmacological tools. Degradation of GABA by GABA-transaminase (GABA-T) was blocked by gamma -vinyl-GABA (GVG) and synthesis of GABA through glutamate decarboxylase (GAD) was suppressed with 3-mercaptopropionic acid (NIPA). We measured miniature GABAergic postsynaptic currents (mIPSCs) in CA3 pyramidal cells using the whole-cell patch clamp technique. 3. Elevated intra-synaptic GABA levels after block of GABA-T resulted in increased mIPSC amplitude and frequency. In addition, tonic GABAergic background noise was enhanced by GVG. Electron micrographs from inhibitory synapses identified by immunogold staining for GABA confirmed the enhanced GABA content but revealed no further morphological alterations. 4. The suppression of GABA synthesis by HPA had opposite functional consequences: mIPSC amplitude and frequency decreased and current noise was reduced compared with control. However, we were unable to demonstrate the decreased GABA content in biochemical analyses of whole slices or in electron micrographs. 5. We conclude that the transmitter content of GABAergic vesicles is variable and that postsynaptic receptors are usually not saturated, leaving room for up-regulation of inhibitory synaptic strength. Diese werden oft mit Humankapitalentwertung, der Signalwirkung von (langen) Erwerbsunterbrechungen und der möglichen Diskriminierung von Frauen auf dem Arbeitsmarkt erklärt. Vernachlässigt wurde in der bisherigen Forschung jedoch die Frage, welchen Einfluss es hat, ob Mütter ihre Erwerbsunterbrechung über die gesetzlich vorgesehene Freistellungsphase hinaus ausdehnen oder nicht. Wir erwarten vor allem für Mütter, die länger als die gesetzlich maximal vorgesehene Freistellungsphase zu Hause bleiben, Lohneinbußen. Daneben untersuchen wir, inwieweit sich die negativen Folgen von Erwerbsunterbrechungen zwischen Müttern, die über unterschiedlich hohes Humankapital in Form von beruflichen Abschlüssen verfügen, und zwischen Ost- und Westdeutschland unterscheiden. Unsere Analyse basiert auf den Daten des SOEP (1984 bis 2011) und kontrolliert Effekte unbeobachteter Heterogenität. Insgesamt zeigen die Ergebnisse, dass es keine generelle und dauerhafte „motherhood wage penalty“ gibt, sondern dass vor allem Unterbrechungen, die über die gesetzlich vorgesehene Phase hinaus gehen, zu niedrigeren Löhnen führen, da sie wahrscheinlich als Signal für eine niedrige Karriereorientierung der Mütter fungieren. This study was conducted to investigate whether LHRH affects apoptosis in ovarian cancer. EFO-21 and EFO-27 ovarian cancer cells were treated with LHRH agonist Triptorelin or with cytotoxic agent Doxorubicin in the absence or presence of Triptorelin. Apoptotic cells were quantified by flow cytometry. Expression of nuclear factor kappa B (NF kappa B) was assessed by RT-PCR and immunoblotting. For determination of Triptorelin-induced NF kappa B activation, cells were transfected with a NF kappa B-secreted alkaline phosphatase reporter gene plasmid (pNF kappa B-SEAP) and cultured for 96 h with or without Triptorelin. The causal relation between Triptorelin-induced NF kappa B activation and Triptorelin-induced protection against apoptosis was investigated using SN50, an inhibitor for nuclear translocation of activated NF kappa B. Apoptosis induction by Triptorelin was never observed. Treatment with Doxorubicin (1 nmol/L) for 72 h increased the percentage of apoptotic cells in EFO-21 and EFO-27 ovarian cancer cell lines to 31% or 34%, respectively. In cultures treated simultaneously with Triptorelin (100 nmol/L), the percentage of apoptotic cells was reduced significantly, to 17% or 18%, respectively (P < 0.001). RT-PCR and immunoblotting experiments showed that NF kappa B subunits p50 and p65 were expressed by ovarian cancer cell lines EFO-21 and EFO-27. When EFO-21 or EFO-27 cells were transfected with pNF kappa B-SEAP and subsequently treated with Triptorelin (100 nmol/L), NF kappa B-induced SEAP expression increased 5.3-fold or 4.7-fold, respectively (P < 0.001). Triptorelin-induced reduction of Doxorubicin-induced apoptosis was blocked by SN50-mediated inhibition of NF kappa B translocation into the nucleus. We conclude that LHRH induces activation of NF kappa B and thus reduces Doxorubicin-induced apoptosis in human ovarian cancer cells. Hingegen gehen Erwerbsunterbrechungen über die reguläre Freistellungsphase hinaus mit deutlich höheren und dauerhaften Verdiensteinbußen einher; dies betrifft knapp 40 Prozent aller in die Untersuchung einbezogenen Mütter. Ein Grund für die erhebliche Verdienstdifferenz könnte in einem negativen Signaleffekt liegen: Aus einer langen Erwerbsunterbrechungen könnten Arbeitgeber auf eine geringere Berufs- und Karriereorientierung der betreffenden Mütter schließen und ihnen daher niedrigere Löhne zahlen. If the break duration falls within the maximum legal length, the deleterious effects on the mother's subsequent working income are only weak and short-lived; but if the break exceeds the maximum legal length, the mother experiences significantly higher and longer-lasting income loss. This affects nearly 40 percent of all mothers included in this study. One reason for this considerable discrepancy may lie in a negative signal effect: After long breaks, employers could reduce mothers to lower professional and career orientations, and therefore pay them lower wages. The aim of this study was to monitor gingival expression of antimicrobial peptides (AMPs) and interleukin-8 (IL-8) in an early gingivitis model. Material and MethodsExperimental gingivitis was allowed to develop in healthy volunteers (n=17). Bleeding on probing (BOP%) and gingival crevicular fluid volume (GCF) were assessed at baseline and day 1, 3, 5, 7 and 14. Expression of AMPs (human beta-defensin-2, hBD-2; CC-chemokine ligand 20, CCL20; psoriasin, pso/S100A7) and IL-8 was analyzed by immunohistochemistry in gingival biopsies. In addition, hBD-2 and IL-8 protein expression was monitored in GCF using the ELISA technology. ResultsExperimental gingivitis gradually developed with an increase in BOP scores and GCF volume over time. In GCF, elevated concentrations of hBD-2 and IL-8 were monitored at day 1, 5 and 7 (p0.0002). Immunohistochemical analysis of gingival sections demonstrated increased staining for hBD-2 at day 3, whereas the CCL20, pso/S100A7, and IL-8 expression was increased at later time points (p<0.05). ConclusionFor the first time, this study showed the time-dependent regulation of AMPs, following clinical signs of experimentally induced gingival inflammation.
