In this context, the establishment of protected areas is a common conservation measure where institutions are put in place and sanctions regarding forest use are enforced. This paper focuses on the practice of sustainable forest management and the associated perspectives of local institutions at the rainforest margins of Lore Lindu National Park (LLNP) in Central Sulawesi, Indonesia. Our case study applies a qualitative social science research approach. Interviews and group discussions with relevant actors such as farmers, village authorities, the National Park authority, and non-governmental organization members were conducted. The Institutional Analysis and Development (IAD) framework served to structure the study and to provide a set of questions to be considered concerning rules, participants, and conservation outcomes. State-induced formal rules are compared with traditional informal rules regarding natural resource use. Our results suggest that the current state-imposed formal rules have not been successfully implemented in the past. Insufficient boundary demarcation, and a lack of congruence between rules and local conditions have been identified as main reasons. Traditional informal rules are rather more respected by local people since they are adapted to traditional use rights and sanctions at the village level. Community conservation agreements (CCAs) are considered a promising tool to mediate between National Park conservation interests and local people's needs integrating traditional informal rules. However, the CCAs implemented in the LLNP area do not address existing differences in perception and behavior of indigenous people and migrants in the area. We argue that this is a central aspect in terms of successful CCA implementation and forest management. Thus, we recommend that the National Park authority should take the cultural diversity of the area seriously into consideration and integrate flexible and distinct socio-cultural strategies into its management processes. Potentially, ecosystems show nonlinear responses to land-use intensification that would open management options with limited ecological losses but satisfying economic gains. However, multidisciplinary studies to quantify ecological losses and socioeconomic tradeoffs under different management options are rare. Here, we evaluate opposing land use strategies in cacao agroforestry in Sulawesi, Indonesia, by using data on species richness of nine plant and animal taxa, six related ecosystem functions, and on socioeconomic drivers of agroforestry expansion. Expansion of cacao cultivation by 230% in the last two decades was triggered not only by economic market mechanisms, but also by rarely considered cultural factors. Transformation from near-primary forest to agroforestry had little effect on overall species richness, but reduced plant biomass and carbon storage by ≈75% and species richness of forest-using species by ≈60%. In contrast, increased land use intensity in cacao agroforestry, coupled with a reduction in shade tree cover from 80% to 40%, caused only minor quantitative changes in biodiversity and maintained high levels of ecosystem functioning while doubling farmers' net income. However, unshaded systems further increased income by ≈40%, implying that current economic incentives and cultural preferences for new intensification practices put shaded systems at risk. We conclude that low-shade agroforestry provides the best available compromise between economic forces and ecological needs. However, its socio-economic implications are under-examined. Analyzing farm-household data from Jambi Province, Sumatra, oil palm adoption is found to have positive consumption and nutrition effects. However, these effects are largely due to farm size expansion that is associated with oil palm adoption. Potential het- erogeneity of effects among oil palm adopters is examined using quantile regressions. Membrane currents evoked by repeated noxious heat stimuli (43-47 degreesC) of 3 s duration were investigated in acutely dissociated dorsal root ganglion (DRG) neurones of adult rats. The heat stimuli generated by a fast solution exchanger had a rise time of 114 +/- 6 ms and a fall time of 146 +/- 13 ms. 2. When heat stimuli were applied to heat-sensitive small (less than or equal to 32.5 mum) DRG neurones, an inward membrane current (I-heat) with a mean peak of 2430 +/- 550 pA was observed (n = 19). This current started to activate and deactivate with no significant latency with respect to the heat stimulus. The peak of I-heat was reached with a rise time of 625 +/- 115 ms. When the heat stimulus tvas switched off I-heat deactivated with a fall time of 263 +/- 17 ms. 3. During constant heat stimulation I-heat decreased with time constants of 4-5 s (inactivation). At the end of a 3 a heat stimulus the peak current was reduced by 44 +/- 5% (n = 19). 4. Current-voltage curves revealed outward rectifying properties of I-heat and a reversal potential of -6.3 +/- 2.2 mV (n = 6). Inactivation was observed at all membrane potentials investigated (-80 to 60 mV); however, inactivation was more pronounced for inward currents (37 +/- 5%) than for outward currents (23 +/- 6%, P<0.05). 5. When neurones were investigated with repeated heat stimuli (3 to 5 times) of the same temperature, the peak current relative to the first I-heat declined by 48 +/- 6% at the 3rd stimulus (n = 19) and by 54 +/- 18% at the 5th stimulus (n = 4; tachyphylaxis). 6. In the absence of extracellular Ca2+ (buffered with 10 mM EGTA) inactivation (by 53 +/- 6%) and tachyphylaxis (by 42 +/- 7% across three stimuli) were still observed (n = 8). The same was true when intracellular Ca2+ was buffered by 10 mw BAPTA (inactivation by 49 +/- 4%, tachyphylaxis by 52 +/- 7% across three stimuli; n=13). Thus, inactivation and tachyphylaxis were mainly independent of intra- and extracellular Ca2+. 7. These results indicate that inactivation and tachyphylaxis of heat-evoked inward currents can be observed in vitro, similar to adaptation and suppression of action potential discharges elicited by comparably fast heat stimuli in vivo. Whereas the voltage dependence of I-heat resembles that of capsaicin-induced membrane currents (I-Caps), the independence of inactivation and tachyphylaxis of I-heat from calcium is in clear contrast to I-heat. A similar difference in calcium dependence of inactivation has been reported between heat-evoked and capsaicin-induced currents through the cloned capsaicin receptor channel VR1. The paper presents the common sampling frame of the socio economic sub-projects. The investigations and data collections intertwine and complement one another. Thus the methodological approach reflects the idea of an interdisciplinary and integrative research approach. Lead by hypotheses we structured our sampling procedure hierarchically. Starting at the household level in the core villages of the research regions we investigate additional local villages. Further we extend the data collections on the regional level with household, village and trader surveys. The national and international levels we reach by stakeholder interviews with governmental and non-governmental experts. However, system technologies are often location specific and characterized by partial adoption and disadoption. Previous studies were often not able to fully explain this, because they mostly relied on farm and household level data, neglecting plot level differences that may be important. We address this limitation, using SRI adoption in Timor Leste as an example. Regression models are specified and estimated to explain the farmers’ decision-making processes. Participation in training programs and household labor availability increase the probability and intensity of adoption, as SRI is knowledge and labor intensive. However, many other household variables are not significant, while plot level characteristics, such as proximity to the homestead, water control capacity, slope, and soil conditions, have more explanatory power. The results suggest that plot level data are important to understand the adoption of system technologies. In Indonesia, the world's leading palm oil producer, in addition to large companies, smallholder farmers are increasingly engaged in the oil palm sector. Smallholder oil palm cultivation may contribute to income gains and socio-economic development. However, land-use decisions by smallholders are not well understood. Without appropriate policies, negative social and environmental consequences can also occur. To improve the knowledge base, we use data about present and past land-use decisions from a survey of farm households in Sumatra. Employing duration models, we analyse the determinants and dynamics of oil palm adoption among smallholders. We find that independently operating farmers are currently driving growth rates in the oil palm sector. Smallholder adoption decisions are mainly attributable to regional and village level factors. While the current adoption primarily occurs outside of contracts, previous contractual ties between companies and other farmers in the same village play an important role for individual decisions. Beyond initial adoption, we also analyse later expansion decisions. While expanding the oil palm area subsequent to initial adoption is common among all types of adopters, those without previous contracts are found to expand significantly faster. We conclude that the concessions the government has allocated to palm oil companies in the past have initiated oil palm adoption in the small farm sector, but that the ensuing land-use dynamics are mostly beyond government control. To enhance the detection frequency of target genes in screens for new natural products, we have combined for the first time DNA-based SIP with the construction of metagenomic libraries. To isolate genes encoding coenzyme B-12-dependent glycerol dehydratases an enrichment of glycerol-fermenting microorganisms from a sediment sample of the Wadden Sea was performed by using glycerol-C-13(3) as sole carbon source. Subsequently, the C-13-labeled DNA was separated from the naturally abundant C-12-DNA by density centrifugation, and used for library generation.
