ARSs have been implicated in autosomal dominant and autosomal recessive human disorders. Autosomal dominant variants in tryptophanyl-tRNA synthetase 1 (WARS1) are known to cause distal hereditary motor neuropathy and Charcot-Marie-Tooth disease, but a recessively inherited phenotype is yet to be clearly defined. Seryl-tRNA synthetase 1 (SARS1) has rarely been implicated in an autosomal recessive developmental disorder. Here, we report five individuals with biallelic missense variants in WARS1 or SARS1, who presented with an overlapping phenotype of microcephaly, developmental delay, intellectual disability, and brain anomalies. Structural mapping showed that the SARS1 variant is located directly within the enzyme's active site, most likely diminishing activity, while the WARS1 variant is located in the N-terminal domain. We further characterize the identified WARS1 variant by showing that it negatively impacts protein abundance and is unable to rescue the phenotype of a CRISPR/Cas9 wars1 knockout zebrafish model. Eine breite, sozialwissenschaftliche Debatte über Probleme, Folgen und Erklärungsfaktoren ist jedoch auf ein valides, reliabel und verallgemeinerbares Verständnis von Politik und Wandel in der deutschen Urheberrechtspolitik angewiesen. Zentral sind in diesem Zusammenhang einerseits aktuelle Fragen über die Durchsetzungskrise aufgrund der allgegenwärtigen, digitalisierten Kunst und Kultur sowie die Kopplung mit einer eventuellen Netz- und Digitalpolitik. Andererseits lassen sich mit dem Punctuated-Equilibrium-Modell (PE) langfristiger, regulatorischer Politikwandel sowie Wandeltrends von Governance- und Regulierungsmodi bisher unbeachtete Prozesse untersuchen. In einer deduktiv-induktiven Dokumentenanalyse von 146 Gesetzen, Verordnungen und ihren Novellierungen von 1870 bis 2019 wird der Politikwandel qualitativ und quantitativ untersucht. Besondere Aufmerksamkeit gilt der transparenten Ausführung und bruchlosen Integration von individueller Kodierung und statistischer Auswertung. Entlang des orthodoxen Konzepts der Policy Dimensionen von Hall und dessen Weiterentwicklung von Howlett, wird ein bottom-up-Vorgehen deskriptiver Wandel- und Politikfelderhebung vorgeschlagen, welches Interdependenzbeziehungen bei Zielen, Instrumenten und Implementationsstrukturen sowie Feedbackbeziehungen einbezieht. Die Kalibrierungsebene wird entlang der Indikatoren des Dichte-Intensitäts-Ansatzes (Knill) vorgenommen, während im zentrale Ziel-Instrumenten-Bereich mit Rückgriff auf Salamon ein Doppel-Item aus Zielgruppen und Aktivitäten entwickelt und erhoben wird. Politikwandel wird in den Kategorien des historischen Institutionalismus und mit den Konzepten Stretching, Patching, Thickening und bricolage eingeführt. Mit Bezug zur Governance-Literatur wird der Wandel von Steuerungsmodi sowie die Einbettung in ein Multi-Level-System untersucht. Empirisch lässt sich dadurch Urheberrechtspolitik als Policy mit ausschließlich Layering-Wandelsprozessen aufgrund beständigen Regelwachstums charakterisieren. Dieser Wandel findet meist nicht punktuell wie 1965, sondern in Wandelsperioden in den 1870er, 1900er sowie den 1930er statt. Seit 1990 hat eine beständige Wandelsperiode begonnen, die einer erwarteten Krisendynamik nur mit Qualifizierung entspricht. Relevanter ist jedoch der Restrukturierungsprozess, der sich seit 1965 und verstärkt seit der Jahrtausendwende abzeichnet: Zu den üblicherweise beachteten Urhebern mit ihren vertraglich handel- und gerichtlich durchsetzbaren Verfügungsrechtem treten neue Zielgruppen der Industrien und Nutzergruppen, neuen Vergütungsziele, finanz- und informationsbasierte sowie prozedurale Instrumente und schließlich neue Implementeure, die Verwertungsgesellschaften. Dadurch wandelt sich das Politikfeld von meta- zu einer erzwungene Selbstregulierung einerseits und in seinem Steuerungsmodus von Wettbewerb hin zu Verhandlungen. Damit Messungen von Urheberrecht nicht idiosynkratisch bleiben, müssen Indizes daher bei Implementeuren, Vertragstypen und statt bei der Regelstrenge bei den regulierten Konstellationen ansetzen. Der Policy-Forschung kann die Erweiterung des PE-Modells sowie ein Bottom-Up-Verständnis der Hauptitems von Public Policy als interdependent und vor allem die Zielkategorie als empirische Frage, ob Zielgruppen oder Probleme im Zentrum stehen, zu formulieren, empfohlen werden."],["dc.description.abstract","Since the digitization and politicization of copyright law, an increased number of reforms could not be classified without contradictions. A broad, social-scientific debate about problems, consequences, and explanatory factors, however, depends on a valid, reliable, and generalizable understanding of politics and change in German copyright policy. Central in this context are, on the one hand, current questions about the enforcement crisis due to the omnipresent, digitized art and culture as well as the coupling with a possible network and digital policy. On the other hand, the Punctuated Equilibrium Model (PE) can be used to examine long-term, regulatory policy change and change trends in governance and regulation modes that have previously been ignored. In a deductive-inductive document analysis of 146 laws, ordinances, and their amendments from 1870 to 2019, the political change is examined qualitatively and quantitatively. Particular attention is paid to the transparent execution and seamless integration of individual coding and statistical analysis. Along the orthodox concept of policy dimensions by Hall and its further development by Howlett, a bottom-up approach of descriptive change and policy field survey is proposed, which includes interdependent relationships in goals, instruments, and implementation structures as well as feedback relationships. The calibration level is carried out along the indicators of the density-intensity approach (Knill), while in the central target instrument area with recourse to Salamon, a double item consisting of target groups and activities is developed and recorded. Political change is narrowed down in the categories of historical institutionalism and with the concepts of stretching, patching, thickening and bricolage. With reference to the governance literature, the change of control modes as well as the embedding in a multi-level system is examined. In this way, copyright policy can be empirically characterized as a policy with exclusively layering change processes due to constant rule growth. This change usually does not take place in one go, as in 1965, but rather in periods of change in the 1870s, 1900s and 1930s. A constant period of change has begun since 1990, which corresponds to an expected crisis dynamic only with qualifications. More relevant, however, is the restructuring process that has been emerging since 1965 and increasingly since the turn of the millennia: In addition to the creator who are usually considered with their contractual and legally enforceable exclusive rights, there are new target groups of industries and user groups, new remuneration targets, financial and information-based as well as procedural instruments and finally new implementers, the collecting societies. As a result, the policy field is changing from meta- to forced self-regulation on the one hand and in its control mode from competition to negotiations. So that measurements of copyright law do not remain idiosyncratic, indices must therefore start with implementers, contract types and, instead of the strictness of rules, with the regulated constellations.
